I grease mine 2 times a year just because, but as long as you do it at every tire change you should be ok, unless your getting some noise from them..

how often should the rearsplines be greased?

How many miles do you want to get out of your bike?

The more often you properly lubricate the less often parts will wear out.

I grease the drive shaft splines (Both ends) the splines and pins on the "Clutch Hub" at least once a year. I also use Honda Moly60 paste as it has a high Molybdenum Disulfide content at an affordable price.

Does this involve removing the rear tire?

Removing the rear wheel, yes. The tire stays on the the wheel, of course.

For my bike, I plan to just grease my shaft splines and splines on the wheels on every rear tire change. When I removed my rear wheel the first time, I still can visibly notice some grease left on the splines after 12K miles. One can probably can apply more grease every 15K miles...I know I can because I applied a little more grease on my splines. However, you really could not go wrong with applying new grease every year...it is just like changing your oil more often will not hurt the bike. Have fun with maintenance.
